Gene expression was compared between disease conditions (CF vs. non-CF) and treatments (TNFα+IL-17 vs. control) as well as the interaction between disease and treatment. Overall design: Primary differentiated airway epithelia were treated with TNFα (10ng/ml) and IL-17 (20ng/ml) for 5 hours, prepared for RNA sequencing, and sequenced on Illumina HiSeq 4000 system using 2x150bp paired end SBS chemistry.

本研究对疾病状态(囊性纤维化(Cystic Fibrosis, CF)与非囊性纤维化)、处理条件(肿瘤坏死因子α(Tumor Necrosis Factor α, TNFα)联合白细胞介素17(Interleukin 17, IL-17)与对照组)下的基因表达水平,以及疾病状态与处理条件的交互作用进行了比较分析。整体实验设计如下:原代分化气道上皮细胞经10ng/ml TNFα与20ng/ml IL-17联合处理5小时后,完成RNA测序文库制备,随后基于Illumina HiSeq 4000测序平台,采用2×150bp双端边合成边测序(Sequencing by Synthesis, SBS)化学方法完成测序。
