We generated PSEN1 mutant macaques by genomic deletion of exon 9 using two guide RNAs targeting the upstream and downstream intron sequences of PSEN1 exon 9, respectively. Here, blood transcriptome sequencing was performed on these 5-month-old control and mutant monkeys to assess the potential impact of familial Alzheimer's disease-associated PSEN1 mutations on juvenile monkeys.
本研究通过两条分别靶向PSEN1基因第9外显子(exon 9)上下游内含子序列的向导RNA(guide RNA),经基因组删除第9外显子的方式构建了PSEN1突变型猕猴。本研究对这批5月龄的对照及突变型猕猴开展了血液转录组测序(blood transcriptome sequencing),以评估与家族性阿尔茨海默病(familial Alzheimer's disease)相关的PSEN1突变对幼年猕猴的潜在影响。
