T-helper (Th) cells actively communicate with adjacent cells by secreting soluble mediators, and yet crosstalk between Th cells and endothelial cells is poorly understood. Placental growth factor (PlGF), originally identified in the placenta, is an angiogenic factor homologous to vascular endothelial growth factor (VEGF)-A. We performed transcriptome analysis to systemically compare the effect of IL6, a key factor in Th cell polarization, and PlGF on T cell functions. Overall design: Conditioned medium containing PlGF (PlGFCM) was obtained from HEK-293 clones overexpressing PlGF2. Mouse splenic CD4+ T cells were stimulated with anti-CD3 antibody and anti-CD28 antibody in the presence of PlGFCM, control conditioned medium (ConCM), or IL6+ConCM for 12 hours.

T辅助(T-helper, Th)细胞可通过分泌可溶性介质与邻近细胞进行主动通信,然而目前对Th细胞与内皮细胞间的相互串扰仍知之甚少。胎盘生长因子(placental growth factor, PlGF)最初在胎盘中被发现,是一种与血管内皮生长因子A(vascular endothelial growth factor, VEGF-A)同源的血管生成因子。本研究通过转录组分析,系统性比较了Th细胞极化关键因子白细胞介素6(interleukin-6, IL6)与PlGF对T细胞功能的影响。实验整体设计:将过表达PlGF2的人胚肾293(human embryonic kidney 293, HEK-293)细胞克隆制备得到含PlGF的条件培养基(PlGFCM)。将小鼠脾脏CD4+ T细胞用抗CD3抗体与抗CD28抗体刺激,并分别置于PlGFCM、对照条件培养基(ConCM)或IL6+ConCM中培养12小时。
