Sela Pass in Tawang district of Arunachal Pradesh, India is situated in the eastern Himalayan global biodiversity hotspot region (Myers et al. 2000) and experiences large temperature fluctuations, a high number of frost and ice days, regular freeze–thaw events and high precipitation. This is a first study for the bacterial diversity of cold adapted regions at such high altitude from North-East India. 16S ribosomal RNA gene clone library from eDNA is generated using pGEMT-easy vector and E.coli DH5α.
印度阿鲁纳恰尔邦(Arunachal Pradesh)达旺县(Tawang district)的色拉山口(Sela Pass)地处东喜马拉雅全球生物多样性热点区域(Myers等,2000),该区域气温波动剧烈、霜冰日数繁多、频繁发生冻融事件且降水充沛。本研究为针对印度东北部此类高海拔寒冷适应区域细菌多样性的首项相关研究。本研究采用pGEMT-easy载体与大肠杆菌(E. coli)DH5α菌株,从环境DNA(environmental DNA,eDNA)中构建得到16S核糖体RNA基因(16S ribosomal RNA gene)克隆文库。
