In Burkholderia pseudomallei, the gene encoding for II2523 contains PAS and GGDEF domains and is responsible for thermoregulation of biofilm formation at 28C and 37C temperatures. A clean deletion mutant of II2523 in Burkholderia pseudomallei Bp82 (a Select Agent exempt strain) and the parental strain were grown as pellicle biofilms in 6-well culture plates at either 28C or 37C in biological triplicate. RNA was extracted from the pellicle biofilms after 24hr growth at the two temperatures and libraries were prepared for sequencing on an Illumina NextSeq500 flow cell. 12 samples in total as follows: deltaII2523 A,B,C -> 28C; deltaII2523 D,E,F -> 37C; Bp82 Parent A,B,C -> 28C; Bp82 Parent D,E,F -> 37C.

在类鼻疽伯克霍尔德菌(Burkholderia pseudomallei)中,编码II2523的基因含有PAS结构域(PAS domain)与GGDEF结构域(GGDEF domain),负责在28℃与37℃条件下对生物被膜的形成进行温度调控。将类鼻疽伯克霍尔德菌Bp82(一种选择代理豁免菌株)的II2523纯合缺失突变株及其亲本菌株,以3次生物学重复的方式,在6孔培养板中于28℃或37℃下培养为浮膜生物被膜(pellicle biofilm)。在两种温度下培养24小时后,从浮膜生物被膜中提取RNA,并在Illumina NextSeq500测序流通池(flow cell)上构建测序文库。本次实验共计12个样本,具体分组如下:deltaII2523 A、B、C组对应28℃;deltaII2523 D、E、F组对应37℃;Bp82亲本株A、B、C组对应28℃;Bp82亲本株D、E、F组对应37℃。
