Knockdown of the antisense noncoding mitochondrial RNAs (ASncmtRNAs) triggers proliferative arrest and apoptotic death of tumor cells from different tissue origins, without affecting normal cells. We aimed to study transcriptomic changes induced by ASncmtRNA knockdown in order to study the mechanisms underlying the effects on tumor cells, specifically in MDA-MB-231 breast cancer cells. MDA-MB-231 cultures were transfected with an antisense oligonucleotide (ASO-1537) targeting the antisense noncoding mitochondrial RNAs (ASncmtRNAs) or with a control ASO (ASO-C) or left untreated, for 24 h. Total RNA was collected and subjected to paired-end Illumina RNA sequencing. Experiments were performed in triplicate.

靶向反义非编码线粒体RNA(antisense noncoding mitochondrial RNAs, ASncmtRNAs)的敲降可诱导不同组织来源肿瘤细胞发生增殖停滞与凋亡性死亡,且不会对正常细胞造成影响。本研究旨在探究ASncmtRNA敲降所引发的转录组变化,以解析其对肿瘤细胞产生效应的潜在分子机制,实验对象为MDA-MB-231乳腺癌细胞。将MDA-MB-231细胞培养物分别用靶向ASncmtRNAs的反义寡核苷酸(ASO-1537)、对照反义寡核苷酸(ASO-C)进行转染,同时设置未处理组,各组处理时长均为24小时。收集总RNA并开展双端Illumina RNA测序,所有实验均设置三次重复。
