Human osteogenically enhanced mesenchymal stem cells were cultured for 5 days on standard gelfoam or gelfoam coated with MSC-derived extracellular matrix Overall design: To address whether mesenchymal stem cell derived extracellular matrix might stimulate the secretion of osteogenic factors by OEhMSCs, we seeded them onto standard gelfoam or ECM-coated gelfoam in vitro and cultured them under standard conditions. After 5 days, we performed transcriptome micro-array analysis to identify genes that were up-regulated >2-fold.
将人成骨增强型间充质干细胞(human osteogenically enhanced mesenchymal stem cells, OEhMSCs)在标准明胶海绵(gelfoam)或经间充质干细胞(mesenchymal stem cell, MSC)来源细胞外基质(extracellular matrix, ECM)包被的明胶海绵上培养5天。
实验整体设计:为探究间充质干细胞来源细胞外基质是否可促进人成骨增强型间充质干细胞分泌成骨因子,我们将该细胞体外接种于标准明胶海绵或经ECM包被的明胶海绵上,并在标准培养条件下进行培养。培养5天后,我们开展转录组微阵列分析,以筛选上调幅度超过2倍的差异表达基因。
