Transcriptonal analysis of Tregs reveal role of LAG3 in stability and function of Tregs in an inflamed CNS Overall design: CD45+ cells or CD4 cells were sorted from CNS cells isolated at peaks of diseases in age and sex matched mice(Lag3KO and WT) in which EAE was induced were subject to Gene Expression analysis using the 10x Genomics Single Cell analysis platform.

调节性T细胞(Tregs)转录组分析揭示LAG3在炎症中枢神经系统(CNS)中调节性T细胞的稳定性与功能中的作用。整体实验设计:从诱导了实验性自身免疫性脑脊髓炎(EAE)的年龄、性别匹配的Lag3基因敲除(Lag3KO)与野生型(WT)小鼠的疾病发作高峰期分离的中枢神经系统细胞中,分选得到CD45阳性细胞或CD4阳性细胞,随后采用10x Genomics单细胞分析平台开展基因表达分析。
