Emergent metastatic heterogeneity of primary tumor cells is restricted by the Ig-transmembrane protein VSIG IV

Our study prompeted us to assign, through spiked single cell RNAseq, metastatic function to transcriptomic cluster of colon cancer cells. Interactions between clones also uncover a restrictive anti metastatic influence that act intrinsically and non-cell automomously. We identify VSIG1 as critical mediator of this influence. 本研究通过加标单细胞RNA测序（spiked single cell RNAseq），明确了结肠癌细胞转录组聚类簇的转移相关功能。克隆间的相互作用还揭示了一种兼具内在调控与非细胞自主双重作用的限制性抗转移效应。本研究鉴定出VSIG1是该效应的关键介导因子。实验整体设计：对感染了携带靶向VSIG1的短发夹RNA（shVSIG1）慢病毒以及空载对照PLKO慢病毒的C11细胞克隆开展RNA测序。
