To elucidate the molecular mechanism involved in increased tolerance to high-salinity stress by the application of HDAC inhibitor named SAHA Overall design: Cassava cultivar TMS6044 was asexually propagated in a plastic pot with MS medium. The 5-cm length shoot top of 3-month-old cassava plantlets were cut and then transferred to liquid MS media without agar. After one week incubation, the plantlets showing root elongation were treated with 100 µM SAHA for 24 h, followed by treatment with 200 mM NaCl for 2 and 24 h. 100 ng of total RNA were used.

实验设计:以木薯品种TMS6044为材料,在含MS培养基的塑料盆中进行无性繁殖。选取3月龄组培苗顶端5 cm长的顶梢剪下,转接至不含琼脂的液体MS培养基中培养。经过1周孵育后,对已出现根伸长的组培苗施加100 μM SAHA处理24小时,随后分别用200 mM NaCl处理2小时和24小时。实验采用100 ng总RNA开展后续分析。
