To compare the effects of RXR inhibition to those of APC depletion on the intestinal epithelium, WT organoids were treated with either HX531 (RXRi) or vehicle (WT) and APCko tumorids were treated with vehicle only. We then performed gene expression profiling analysis using data obtained from mRNA-seq of the indicated organoid lines. Wild type intestinal organoids (WT), RXRi and ApcKO (A) were cultured in media with either HX531 or vehicle before collecting for RNA extraction.

为对比RXR抑制(RXR inhibition)与APC敲除(APC knockout, APCko)对肠道上皮的影响,本研究将野生型(Wild Type, WT)类器官分别施以HX531(RXR抑制剂,RXRi)或其溶剂对照处理(记作WT组),同时仅对APCko肿瘤类器官施以溶剂对照处理。随后,我们利用指定类器官株系的mRNA测序(mRNA-seq)数据开展基因表达谱分析。野生型肠道类器官(WT)、RXRi处理组与APCko组(标注为A)在添加HX531或溶剂对照的培养基中培养,收集样本后进行RNA提取。
