To understand the ontogeny and longevity of tissue-resident macrophages in nonhuman primates (NHPs), we employ a model of autologous hematopoietic stem progenitor cell (HSPC) transplantation with HSPCs genetically modified to be marked with clonal barcodes, allowing for subsequent analysis of clonal ontogeny. Macrophages from animals that had been transplanted with barcode- and GFP-labeled stem cells were compared with non-GFP macrophages and macrophages from control, unmanipulated animals.
为探究非人灵长类动物(nonhuman primates, NHPs)体内组织驻留巨噬细胞的个体发生与存活寿命,本研究采用经基因修饰以克隆条形码标记的自体造血干祖细胞(hematopoietic stem progenitor cell, HSPC)移植模型,可用于后续的克隆起源分析。本研究将接受条形码与绿色荧光蛋白(green fluorescent protein, GFP)标记干细胞移植的动物来源巨噬细胞,与非GFP标记巨噬细胞及未接受任何实验操作的对照动物来源巨噬细胞进行了对比分析。
