Single cell RNA-Seq was performed on ID1 ID3 null endothelial cells to determine the effects of ID gene loss on endothelial cell maintainence and growth. Overall design: 20,000 SECs and type H vessels from CDH5-CreER ID1+/+ ID3+/- or CDH5-CreER ID1-/- ID3-/- mice were sorted using FACs and subjected to single cell RNA-Seq.

本研究针对ID1、ID3双功能缺失内皮细胞开展单细胞RNA测序(Single cell RNA-Seq),旨在明确ID基因缺失对内皮细胞稳态维持与增殖的影响。 实验设计:从携带CDH5-CreER重组酶系统、基因型分别为ID1+/+ ID3+/- 与ID1-/- ID3-/-的小鼠中,分选出20000个窦状内皮细胞(SECs)及H型血管,经荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACs)处理后,对上述样本进行单细胞RNA测序。
