We performed ChIP-seq for H3K4me3, H3K27ac, H3K4me1, H3K27me3, H3K36me3, H3K9me3, and H4K20me3 to characterize chromatin states and investigated SOX2 deposition in asynchronous and mitotic cells. Overall design: Examination of H3K4me3, H3K27ac, H3K4me1, H3K27me3, H3K36me3, H3K9me3, and H4K20me3 deposition in asynchronous and mitotic mESCs. Examination of SOX2 binding in in asynchronous and mitotic mESCs.
本研究针对H3K4me3、H3K27ac、H3K4me1、H3K27me3、H3K36me3、H3K9me3及H4K20me3开展染色质免疫共沉淀测序(Chromatin Immunoprecipitation Sequencing, ChIP-seq)以表征染色质状态,并探究了异步培养与有丝分裂期细胞中SOX2的染色质结合与沉积特征。整体实验设计:检测异步培养及有丝分裂期小鼠胚胎干细胞(mouse embryonic stem cells, mESCs)中上述组蛋白修饰的分布;同时检测该两类细胞中SOX2的染色质结合情况。
