Protection against skin aging is important for not only facial appearance but also in the terms of biological defence. In this study, the mechanism of human skin aging was identified by in vitro multi-omics analysis. Replicative stress was introduced into human dermal fibroblasts, HFF-1, as a senescent cell model. RNA-seq, ATAC-seq, and ChIP-seq (histone H3 lysine 27 (H3K27Ac)) were performed on different passages of HFF-1.

抵御皮肤衰老不仅对维持面部外观至关重要,在生物防御层面同样具有关键意义。本研究通过体外多组学分析,阐明了人类皮肤衰老的作用机制:通过向人类皮肤成纤维细胞HFF-1中引入复制应激,构建其衰老细胞模型;随后针对不同传代次数的HFF-1细胞,开展了RNA测序(RNA-seq)、转座酶可及性测序(ATAC-seq)以及组蛋白H3赖氨酸27乙酰化染色质免疫沉淀测序(ChIP-seq,H3K27Ac)实验。
