To further investigate the mechanism of SphK1/2 on oocyte maturation, we established Sphk1/2gc -/- and WT mice. We then performed transcriptomic analysis on the MGCs and cumulus cells of Sphk1/2gc-/- and WT mice at 2 h post-hCG. Comparative gene expression profiling analysis of RNA-seq data for the MGCs and cumulus cells of Sphk1/2gc-/- and WT mice at 2 h post-hCG.

为深入探究鞘氨醇激酶1/2(SphK1/2)调控卵母细胞成熟的作用机制,我们构建了生殖细胞特异性Sphk1/2敲除(Sphk1/2gc-/-)小鼠与野生型(Wild Type, WT)对照小鼠。随后,我们于人绒毛膜促性腺激素(human chorionic gonadotropin, hCG)处理后2小时,采集上述小鼠的壁层颗粒细胞(mural granulosa cells, MGCs)及卵丘细胞并开展转录组学分析。同时,针对hCG处理后2小时的Sphk1/2gc-/-与WT小鼠的MGCs及卵丘细胞的RNA测序(RNA-seq)数据,我们进行了比较基因表达谱分析。
