Expression profiling of luminal and glandular epithelia in the neonatal and adult mouse uterus

Laser capture microdissection coupled with microarray genes expression analysis were utilized in order to elucidate the regulatory networks active in epithelial cells of the neonatal and adult mouse uterus. Genes predominantly expressed in the glandular epithelium (GE) in the neonatal or adult uterus were determined by comparing transcripts in the isolated samples of luminal epithelium (LE) and GE from postnatal day (PD) 10, day of pseudopregnancy (DOPP) 2.5 or DOPP 3.5 uteri (n=2/cell type/day).

本研究采用激光捕获显微切割(Laser capture microdissection)联合微阵列基因表达分析技术,旨在阐明新生及成年小鼠子宫上皮细胞内的活跃调控网络。通过对比出生后第10天(PD 10)、假孕第2.5天(DOPP 2.5)或假孕第3.5天(DOPP 3.5)小鼠子宫的腔上皮(LE)与腺上皮(GE)分离样本中的转录本,最终确定了在新生或成年小鼠子宫腺上皮中优势表达的基因;实验中每组细胞类型与对应时间点设置2个生物学重复(n=2/cell type/day)。
