In this study, we evaluated age-related alterations in the bacterial and fungal communities which are present in the ileo-cecal lymph nodes of suckling and newly weaned piglets. The study was conducted in two replicate batches. Piglets received only sow milk or additional creep feed from day of life (DoL) 10. Piglets were weaned on DoL28. Piglets were sacrified on DoL 7, 14, 21, 28, 31 and 35 and the ileo-cecal lymph nodes were collected (n=5/diet/sex/day). The RNA was isolated from the lymph nodes which was transcribed into complementary DNA (cDNA). The cDNA was sequenced using Illumina chemistry. The V3-V4 hypervariable regions of bacterial 16S rRNA gene and the ITS2 amplicon were amplified using a 250 bp read length paired-end protocol (Novogene) on an Illumina Novaseq 6000 platform, including the library preparation (NEBNext Ultra II DNA Library Prep Kit) and sequencing. 本研究旨在评估哺乳仔猪与刚断奶仔猪回盲淋巴结中细菌及真菌群落的年龄相关性变化。本实验设置两个重复批次。仔猪于出生后日龄(day of life,DoL)10时,仅饲喂母猪乳汁,或额外补充教槽料;仔猪于DoL28时完成断奶。分别于DoL7、14、21、28、31及35天对仔猪实施安乐死,采集回盲淋巴结样本,每组按日粮、性别、采样日龄分组,每组样本量为5(n=5)。从淋巴结样本中提取总RNA,将其反转录为互补脱氧核糖核酸(complementary DNA,cDNA)。采用Illumina测序技术对cDNA进行测序:针对细菌16S rRNA基因V3-V4高变区及ITS2扩增子,采用250bp读长的双端测序方案,由诺禾致源(Novogene)在Illumina NovaSeq 6000平台完成测序,实验涵盖文库构建环节,所用试剂盒为NEBNext Ultra II DNA Library Prep Kit。本生物项目对应ITS2扩增子测序数据,细菌16S序列数据可在PRJNA1050750项目编号下获取。
