Osteoarthritis (OA) is the most common equine joint disease characterised by cartilage degradation and changes to other joint tissues severely effecting welfare and performance leading to early retirement. It results in substantial morbidity and mortality, but our understanding of the pathophysiological mechanisms involved is limited. There are no disease modifying therapeutics available, with OA medication offering symptomatic relief only. Early diagnosis is thus important, as substantial joint pathology is usually present at the time of clinical diagnosis. Thus, biomarkers of early-stage OA are actively sought, and among these, extracellular vesicles (EVs) are emerging as potential candidates. Here we use a SWATH MS approach to profile the protein cargo of EVs derived from equine plasma and synovial fluid.

骨关节炎(Osteoarthritis, OA)是最常见的马类关节疾病,以软骨退变及其他关节组织病变为特征,严重影响马匹的健康与运动表现,进而导致其提前退役。该病可造成显著的发病率与病死率,但目前学界对其相关病理生理机制的认知仍较为有限。当前尚无可用的疾病修饰治疗药物,骨关节炎的治疗仅能实现症状缓解。因此早期诊断至关重要,因为临床确诊时往往已存在较为显著的关节病理改变。故而学界正积极探寻早期骨关节炎的生物标志物,其中细胞外囊泡(extracellular vesicles, EVs)逐渐成为潜在候选对象。本研究采用SWATH质谱(SWATH MS)技术,对源自马血浆与滑液的细胞外囊泡所携带的蛋白质组进行谱分析。
