Hybridization and study was conducted to measure the impact of short-term sugar deprivation, on an acclimatization time-scale, imposed to plantlet transferred from in vitro to ex vitro culture conditions. We thus submitted in vitro tomato plantlets to sugar deprivation for 24-h in order to better understands the impact of sucrose retrieval upon ex vitro acclimatization. A total of six microarrays were used for this experiment. More precisely, three biological replicates represented by individually grown in vitro plantlet from both treatments were hybridized. Dye swap was conducted as technical replicate in order to minimize dye bias.
本实验开展杂交与相关研究,旨在探究将幼苗从离体(in vitro)培养条件转接至试管外(ex vitro)驯化培养环境后,短期糖剥夺对其驯化进程的影响。为此,我们将离体培养的番茄幼苗施以24小时糖剥夺处理,以更深入解析蔗糖恢复供给对试管外驯化过程的作用效果。本实验共使用6张基因芯片(microarrays)。具体而言,本实验针对两种处理组,各选取3个生物学重复(biological replicates)样本——即单独培养的离体番茄幼苗——进行杂交实验。同时通过染料互换(dye swap)设置技术重复,以最小化染料偏倚带来的实验误差。
