Using ACE-seq we investigated single-base resolution hydroxymethylomes of sea urchin, lancelet and zebrafish during development. ACE-seq was perfomred on (i) 24 hpf, 48 hpf, 72 hpf embryos and adult tissue (tube feet) of purple sea urchin in biological replicates; (ii) 36 hpf, larvae (60-70 hpf), juvenile (3-4mo) and adult tissue (liver) of european lancelet in biological replicates (larvae and juvenile stages); (iii) zebrafish 24 hpf embryo.
本研究采用ACE-seq技术,对发育进程中的海胆、文昌鱼与斑马鱼的单碱基分辨率羟甲基化组(hydroxymethylomes)开展检测分析。具体样本与实验设置如下:
(i) 紫色海胆:24 hpf(受精后小时,hours post fertilization)、48 hpf、72 hpf胚胎,以及成体管足组织,所有样本均设置生物学重复;
(ii) 欧洲文昌鱼:36 hpf胚胎、幼虫期(60-70 hpf)、幼体期(3-4月龄)以及成体肝脏组织,其中幼虫与幼体阶段设置生物学重复;
(iii) 斑马鱼:24 hpf胚胎。
