In this study, we adopted a new experimental idea to dynamically discover HHT drug resistance by constructing HHT resistant cell lines with a gradually increasing resistance index(RI). Furtherly, we did RNA-sequence among HHT sensitive cells and the cells with a gradually increasing RI. Through the biological function and pathway analysis of the RNA-seq data reveal the mechanism of HHT resistantce.

本研究采用全新实验思路，通过构建耐药指数（Resistance Index，RI）逐步升高的高三尖杉酯碱（Homoharringtonine，HHT）耐药细胞系，实现对HHT耐药性的动态解析。进一步地，本研究对HHT敏感细胞与上述耐药指数逐步升高的耐药细胞开展RNA测序（RNA-seq）；通过对测序数据进行生物学功能与通路分析，阐明了HHT耐药的分子机制。
