Purpose: The goal of this study was to compare the mRNA transcriptomes of transgenic pigs and wild-type pigs for five organs (liver,longissimus dorsi,psoas major,subcutaneous fat and retroperitoneal fat). Methods: Total RNA was extracted from longissimus dorsi, psoas major, subcutaneous fat, retroperitoneal fat and liver using TRIZOL reagent (Life Technologies, Beijing, China), and each tissue has three biological replicates. For RNA sequencing, approximately 2 ug total RNA of each sample was used to construct transcriptome libraries following the manufacture’s standard procedures. Libraries were sequenced via Illumina HiSeqTM 4000, and 100 bp paired-end reads were generated. Results: We detected 18,051 protein coding mRNA transcripts across 30 samples . Conclusions: Human FST induced transcriptome changes in liver, Five organs (liver, longissimus dorsi, psoas major, subcutaneous fat and retroperitoneal fat) mRNA profiles of transgenic pigs and wild type pigs were generated by deep sequencing, in triplicate using Illumina HiSeqTM 4000.

研究目的:本研究旨在比较转基因猪与野生型猪的5种器官(肝脏、背最长肌、腰大肌、皮下脂肪及腹膜后脂肪)的mRNA转录组。实验方法:采用TRIZOL试剂(Life Technologies公司,中国北京)从背最长肌、腰大肌、皮下脂肪、腹膜后脂肪及肝脏中提取总RNA,每份组织设置3个生物学重复。RNA测序环节,取每份样本约2 μg总RNA,按照厂商标准流程构建转录组文库;文库通过Illumina HiSeqTM 4000完成测序,获得100 bp双端读段。实验结果:本研究在30个样本中共检测到18051个蛋白编码mRNA转录本。研究结论:人源FST可诱导肝脏组织的转录组发生改变;通过Illumina HiSeqTM 4000进行深度测序并设置3次生物学重复,成功获取了转基因猪与野生型猪5种器官(肝脏、背最长肌、腰大肌、皮下脂肪及腹膜后脂肪)的mRNA表达谱。
