PDGF启动子下游插入人a-synuclein A53T基因,显微注射获得脑特异表达a-synuclein A53T的转基因大鼠。常染色体显性遗传家族性帕金森病(PD)与α-synuclein的错义突变型A53T相关。该转基因大鼠表现为中脑黑质酪氨酸羟化酶阳性神经元数目显著减少,平衡能力下降,步态障碍,可以用于帕金森病的研究。

By inserting the human α-synuclein A53T gene downstream of the PDGF promoter, transgenic rats with brain-specific expression of α-synuclein A53T were generated via microinjection. Autosomal dominant familial Parkinson's disease (PD) is associated with the missense mutation A53T of α-synuclein. These transgenic rats show a significant reduction in the number of tyrosine hydroxylase-positive neurons in the substantia nigra pars compacta of the midbrain, accompanied by impaired balance and gait disturbances, making them applicable for Parkinson's disease research.
