Contains Figure S1. FACS analysis of adipose tissue stromal vascular cells. Male C57BL6/J mice, at 5–6 weeks of age, were fed a high-fat diet (HFD) and treated with metformin (Met, 150 mg/kg/d, in phosphate-buffered saline (PBS)) or PBS for the last 4 weeks of HFD feeding (n = 4–6). After the feeding/treatment regimen, stromal vascular cells (SVC) were isolated from epididymal fat pads and subjected to FACS analysis. (A) SVC were included for FACS analyses. (B) SVC (without staining) were analyzed for APC and FITC. (C) SVC (with staining) were analyzed for F4/80 (FITC) and CD11b (APC) expression.

本数据集包含补充图S1:脂肪组织基质血管细胞的流式细胞术分析(FACS,Fluorescence Activated Cell Sorting)。实验对象为5~6周龄的雄性C57BL6/J小鼠,先予以高脂饮食(HFD,High-Fat Diet)喂养;在高脂喂养的最后4周,分别给予二甲双胍(Met,Metformin,150 mg/kg/d,溶于磷酸盐缓冲液(PBS,Phosphate-Buffered Saline))或等量磷酸盐缓冲液处理,每组小鼠数量为4~6只(n=4~6)。完成喂养与给药方案后,从附睾脂肪垫中分离基质血管细胞(SVC,Stromal Vascular Cells)并进行流式细胞术分析:(A) 选取基质血管细胞用于流式细胞术分析;(B) 对未染色的基质血管细胞检测APC与FITC通道的荧光信号;(C) 对已染色的基质血管细胞,检测其F4/80(标记于FITC通道)与CD11b(标记于APC通道)的表达情况。(PDF)
