Spink5 constitituve knock-out mice mimic the disease features of Netherton syndrome but die within several hours after birth due to severe skin barrier defect. To characterize the skin barrier and skin inflammation phenotype of this mouse model at the molecular level, we collected back skin samples from mice shortly after their birth. Overall design: Comparative gene expression profiling analysis of RNA-seq data for Spink5 constitutive knock-out newborn mice (n=4) and control (wild-type) newborn mice (n=4).

Spink5组成型基因敲除小鼠(Spink5 constitutive knock-out mice)可模拟内瑟顿综合征(Netherton syndrome)的疾病表型,但因严重皮肤屏障缺陷于出生后数小时内死亡。为从分子水平解析该小鼠模型的皮肤屏障与皮肤炎症表型,我们于小鼠出生后不久收集其背部皮肤样本。实验整体设计:对4只Spink5组成型基因敲除新生小鼠(n=4)与4只野生型对照新生小鼠(n=4)的RNA测序(RNA-seq)数据开展比较基因表达谱分析。
