To study the role of tet3 and 5-hydroxymethylation in mouse intestine homeostasis our objectives are: To determine the cell types that show a 5hmC reduction level in the absence of Tet3. Identify the role of Tet3 on transcriptional regulation in the small intestinal epithelium. Define the physiological functions impaired in Tet3 -/- small intestine epithelium. Single cell RNA-seq analysis of control and Tet3 knockout mice. The Control data is split in 6 fastq files. The Tet3 knockout data is split into 6 fastq files.
本研究旨在探究Tet3(Tet3)与5-羟甲基化(5-hydroxymethylation)在小鼠肠道稳态中的作用,具体目标如下:其一,鉴定在Tet3缺失条件下呈现5hmC水平降低的细胞类型;其二,明确Tet3在小肠上皮转录调控中的功能;其三,阐明Tet3纯合敲除(Tet3 -/-)小鼠小肠上皮中受损的生理功能。本研究对对照组与Tet3敲除小鼠开展单细胞RNA测序(single cell RNA-seq)分析,其中对照组数据被拆分为6个FASTQ文件,Tet3敲除组数据同样被拆分为6个FASTQ文件。
