Whole transcriptome shotgun sequencing (RNA-seq), a high-throughput transcriptome sequencing technology based on the next generation sequencing technology. Through RNA-seq, A large number of mRNA sequences can be obtained quickly and at a lower cost by assembling the huge amounts of sequencing reads, and this is particularly important for species lacking genomic information, such as jojoba. At the same time, by counting the number of reads mapping to specific gene sequences in the genome, RNA-seq can provide the accurate expression level of all genes in a particular tissue or under specific treatment condition readily. Numerous studies using RNA-seq technique have been conducted to establish high-quality reference transcriptome sequence , identified key enzymes in metabolic pathways related to the synthesis of phytochemical constituents, or identify differentially expressed genes under environmental stress in crops and many plant species with high economic, ornamental, or ecological value. However, the transcriptome sequencing of jojoba has not been reported by far, despite its important economic and ecological value.In the present study, we established a general jojoba transcriptome dataset by using RNA-seq.
全转录组鸟枪测序(RNA-seq)是一种基于下一代测序技术的高通量转录组测序技术。借助RNA-seq技术,可通过组装海量测序读段,快速且低成本地获取大量mRNA序列;这对于缺乏基因组信息的物种(如西蒙得木(jojoba))而言尤为关键。同时,通过统计比对至基因组中特定基因序列的读段数量,RNA-seq可便捷地获取特定组织或特定处理条件下所有基因的精准表达水平。目前已有众多研究借助RNA-seq技术,为诸多具有高经济、观赏或生态价值的作物与植物物种构建了高质量的参考转录组序列,鉴定出与植物化学成分合成相关的代谢通路关键酶,或是筛选出环境胁迫下的差异表达基因。然而,尽管西蒙得木具备重要的经济与生态价值,但其转录组测序研究迄今尚未见报道。本研究借助RNA-seq技术,构建了一套通用的西蒙得木转录组数据集。
