To identify transcriptional alterations in subcutaneous human white adipose tissue of post-obese subjects, global gene expression measurements were performed. Three groups, obese before and after bariatric surgery as well as never-obese controls, were compared to dissect candidate genes. Overall design: Based on high quality total RNA isolated from human subcutaneous white adipose tissue, gene expression profiles were generated using Affymetrix Human Gene 1.1 ST arrays (Affymetrix, Inc., Santa Clara, CA). Obese subjects before (n=16) and after bariatric surgery (n=16) as well as never-obese controls (n=16) were included in the study. Data were preprocessed with the Robust Multichip Average method.
为探究既往肥胖受试者皮下白色脂肪组织的转录组变化,本研究开展了全基因表达检测。本研究设置三组样本进行比较以解析候选基因,分别为减肥手术前后的肥胖受试者与从未肥胖的健康对照组。
实验总体设计:基于从人皮下白色脂肪组织中分离得到的高质量总RNA,本研究采用Affymetrix人类基因1.1 ST芯片(美国加利福尼亚州圣克拉拉市Affymetrix公司)构建基因表达谱。本研究共纳入三组受试者:减肥手术前肥胖受试者(n=16)、减肥手术后的既往肥胖受试者(n=16)以及从未肥胖的对照组受试者(n=16)。实验数据采用稳健多数组平均(Robust Multichip Average)方法进行预处理。
