ChIP-seq to map the binding sites for CTCF and cohesin subunit Rad21 in the naive mES cells (46C cell line grown in the 2i/LIF condition) and in the neural stem cells (derived from the 46C ES cells using the mono-layer differentiation protocol, grown in the N2B27 medium these cells are Nestin+). The naive mES cells were grown in two different media (fetal bovine serum, FBS and 2i/LIF culture - naive pluripotency conditions) as detailed in the growth protocols.
本数据集通过染色质免疫共沉淀测序(ChIP-seq)技术,定位CCCTC结合因子(CTCF)与黏连蛋白亚基Rad21在两类细胞中的全基因组结合位点:一类为采用46C细胞系、于2i/LIF培养条件下培养的幼稚型小鼠胚胎干细胞(naive mES cells);另一类为通过单层分化方案由46C胚胎干细胞诱导获得、并在N2B27培养基中培养的神经干细胞(该细胞呈巢蛋白(Nestin)阳性)。前述幼稚型小鼠胚胎干细胞分别采用胎牛血清(FBS)与2i/LIF幼稚多能性两种培养基进行培养,具体培养细节详见对应的生长实验方案。
