We report the genome-wide effects of KAP1 loss on the transcriptome, the chromatin state, and on recruitment of various components of the transcription machinery in the colon colorectal cancer cell line HCT116. 本研究报道了结直肠癌细胞系HCT116中,KAP1缺失对转录组、染色质状态,以及转录机器各组分招募情况的全基因组影响。整体实验设计:在对照组(Ctrl)与KAP1敲除(KO)细胞中,分别检测总RNA聚合酶II(Pol II)及其磷酸化形式、CDK9、NELFE的全基因组结合图谱;检测两组细胞的新生RNA与稳态RNA表达水平;检测两组细胞的核小体占位图谱(微球菌核酸酶测序,MNase-seq)。
