To investigate age-dependent molecular changes in the cortex and hippocampus of Cyfip2 Arg87Cys knock-in mice, we conducted transcriptome analysis at postnatal weeks 1, 7, 14, 28. Overall design: We dissected the frontal cortex and hippocampal regions from both hemispheres of each mouse. WT and Cyfip2 Arg87Cys knock-in mice were littermates, with four mice (replicates) prepared for each age-genotype group. After dissection, tissue samples were stored in RNAlater at 4°C overnight and then at -80°C before being processed further for RNA extraction.
为研究Cyfip2 Arg87Cys敲入小鼠大脑皮层与海马体的年龄依赖性分子变化,我们于小鼠出生后第1、7、14、28周开展了转录组分析。实验设计概况:我们从每只小鼠的双侧大脑分离得到额叶皮层与海马体组织。野生型(Wild Type,以下简称WT)与Cyfip2 Arg87Cys敲入小鼠均为同窝仔鼠,每个年龄-基因型组设置4只小鼠作为生物学重复。组织分离完成后,样本先置于RNAlater固定液中4℃静置过夜,随后转移至-80℃保存,待后续进行RNA提取实验。
