We collected serial sputum samples from TB patients during antibiotics treatment. Three sputum samples were collected at each time points of week 0, 2, 4, 6, 8, 16 and 24. All the isolates were performed both L-J and MGIT culture. All the colonies on L-J plate were scraped and extracted DNA for whole genome sequencing. If the sputum sample was L-J culture negative but MGIT culture positive, the MGIT culture was used for whole genome sequencing. The average sequencing depth was ~1000X.
本研究收集了抗生素治疗期间结核病(Tuberculosis, TB)患者的系列痰液样本。在第0、2、4、6、8、16及24周的各时间点,均采集3份痰液样本。所有分离菌株均同步开展L-J培养基与MGIT培养。刮取L-J平板上的全部菌落并提取DNA,用于全基因组测序(whole genome sequencing, WGS)。若某痰液样本的L-J培养结果为阴性但MGIT培养结果为阳性,则以MGIT培养物进行全基因组测序。测序平均深度约为1000×。
