In this study we investigated the community structure of groundwater bacteria in two superimposed limestone aquifer assemblages in the Hainich Critical Zone Exploratory (Thuringia/Germany). Samples were obtained from 8 different wells along a groundwater observation transect with sampling depth ranging from 12 to 88 m depth. Community structure was assessed by amplicon sequencing using the Illumina MiSeq platform, targeting the V3-V5 region of the bacterial 16S rRNA gene. Sequence analysis was performed for three different time points (August 2014, August 2015, November 2015). In addition, we analysed the community structure of nirS-type denitrifiers in the same groundwater samples obtained in August 2014 targeting nirS genes by Illumina MiSeq.

本研究针对德国图林根州海尼希临界带勘探区(Hainich Critical Zone Exploratory)内的两套叠置石灰岩含水层系统中的地下水细菌群落结构展开了调查。研究沿一条地下水观测断面,从8口不同井位采集了样本,采样深度范围为12至88米。我们采用Illumina MiSeq平台,以细菌16S rRNA基因的V3-V5可变区为靶标,通过扩增子测序(amplicon sequencing)解析群落结构,并分别在2014年8月、2015年8月、2015年11月三个时间点开展序列分析。此外,针对2014年8月采集的同一批地下水样本,我们以nirS基因为靶标,采用Illumina MiSeq平台分析了nirS型反硝化细菌(nirS-type denitrifiers)的群落结构。
