Intrapericardial administration of secretomes from endometrial-derived mesenchymal stromal cells: effects on immune-related genes in a porcine model of myocardial infarction

Using a porcine model of acute myocardial infarction, we explored the potential effect of secretomes from endometrial-derived mesenchymal stromal cells (endMSCs) and from IFNγ/TNFα-primed endMSCs on immune-related genes in the infarcted tissue. Infarcted tissues from pigs administered with placebo (n = 4), secretomes from endometrial mesenchymal stromal cells (S-enMSCs) (n = 4), and secretomes from IFNγ/TNFα-primed endometrial derived mesenchymal stromal cells (S-enMSCs*) were collected at 7 days post-therapy for RNA extraction and Affymetrix microarrays. Differentially gene expression analysis between biogroups were performed and immuno-related genes were determined.
本研究以急性心肌梗死猪模型为对象,探究子宫内膜来源间充质基质细胞(endometrial-derived mesenchymal stromal cells, endMSCs)及其经干扰素γ(IFNγ)/肿瘤坏死因子α(TNFα)预处理的endMSCs所分泌的分泌组,对梗死组织中免疫相关基因的潜在影响。本研究设置三组实验组:安慰剂给药组(n=4)、子宫内膜间充质基质细胞分泌组(S-enMSCs,n=4)以及经IFNγ/TNFα预处理的子宫内膜来源间充质基质细胞分泌组(S-enMSCs*,n=4);于治疗后7天采集各组梗死组织,用于RNA提取与Affymetrix基因芯片检测。随后对各组生物组开展差异基因表达分析,并鉴定免疫相关基因的表达情况。
