Micro-C performed on U2OS cells without or with NFIB depletion at G1/S phase and S phase to investigate the effect of NFIB on higher-order chromatin structure during DNA replication. U2OS cells without or with NFIB depletion at G1/S phase and S phase were synchronized at G1/S boundary by double-thymidine block and followed by release for 4 h to allow cells entering S phase, respectively. Micro-C experiments of those cells in each treatment were biologically repeated twice.

本研究针对G1/S期与S期的U2OS细胞(分设NFIB敲低(NFIB depletion)与未敲低组)开展微-C测序(Micro-C)实验,以探究NFIB在DNA复制过程中对高阶染色质结构的影响。我们通过双胸腺嘧啶阻滞法将上述细胞同步化于G1/S交界点,随后分别释放培养4小时以促使细胞进入S期。各处理组的Micro-C实验均完成了两次生物学重复。
