Understanding underlying mechanisms responsible for the differentiation and stemness of mouse mammary progenitor cells are paramount to understanding the heterogeneity of mammary development and tumorigenesis. Consequently, studies have isolated and characterized mouse mammary progenitor cell lines such as: COMMA-1D, NSP2 and SP3, but have not parsed the various transcriptomes that play a role in progenitor differentiation and mammary gland development. Overall design: Using microarray technologies, we analyzed transcriptomes, from Comma Db clonal derivatives, SP3 and NSP2, during different mouse mammary growth phases: In-vitro, In-vivo 5 weeks, and In-vivo 12 weeks.
阐明调控小鼠乳腺祖细胞分化与干性(stemness)的内在机制,是解析乳腺发育异质性及肿瘤发生过程的核心要务。此前已有研究分离并鉴定了COMMA-1D、NSP2与SP3等小鼠乳腺祖细胞系,但尚未针对参与祖细胞分化及乳腺发育过程的各类转录组(transcriptome)展开系统解析。实验设计:本研究借助微阵列技术(microarray),对Comma Db克隆衍生株、SP3及NSP2细胞系,在小鼠乳腺发育的不同生长阶段——体外培养、体内5周龄、体内12周龄——开展转录组分析。
