In this study, fluorescence microscopy was utilized to observe the growth behaviors under self-incompatible and self-compatible conditions. Scanning electron microscopy (SEM) was employed to analyze the morphological and structural differences between pollen grains and stigmas. Ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was adopted to identify metabolites, and the metabolic mechanism underlying the occurrence of self-incompatibility in *Plumbago auriculata* was finally uncovered.
