RNA-seq analysis was performed in an adult T-cell leukemia/lymphoma cell line (TL-Om1) to analyze gene expression changes after genetic overexpression or knockdown of TP73. For overexpression study, lentiviral infection was carried out on TL-Om1 cells to overexpress TAp73 or ∆Np73 under doxycycline (dox)-inducible system. For knockdown study, lentiviral infection was carried out on TL-Om1 cells to stably express dCas9 and dox-inducible sgRNA targeting TP73 locus. Total RNA was harvested 3 days after dox induction and RNA-seq analysis was performed.

本研究针对成人T细胞白血病/淋巴瘤细胞系TL-Om1开展RNA测序(RNA-seq)分析,以解析TP73基因过表达或敲低后细胞的基因表达变化。过表达实验中,通过慢病毒感染TL-Om1细胞,在多西环素(doxycycline,dox)诱导表达系统中过表达TAp73或∆Np73。敲低实验中,采用慢病毒感染TL-Om1细胞,使其稳定表达dCas9以及靶向TP73基因座的多西环素诱导型单导RNA(sgRNA)。于多西环素诱导3天后收集总RNA,随后开展RNA测序分析。
