Gene expression profiling was performed on tissue samples via a complementary DNA-mediated annealing, selection, extension, and ligation (DASL) assay (Illumina). 1536 were selected for profiling. 210 prostate cancer samples were used.Random forests classifcation was applied to the DASL data, and a set of genes was selected for further development. Total RNA was isolated from prostate cancer prostatecomy samples, purified, cDNA generated, and DASL performed.

本研究通过互补DNA介导的退火、选择、延伸及连接(DASL)检测技术(Illumina公司)对组织样本开展基因表达谱分析。共选取1536个靶标进行表达谱分析。本次研究共使用210份前列腺癌样本。针对DASL检测数据,采用随机森林分类算法进行分析,筛选出一组基因用于后续开发。从前列腺切除术获取的前列腺癌组织样本中提取总RNA,经纯化后合成互补DNA,随后开展DASL检测。
