Supplementary Dataset 1 - Description
By means of bioinformatics analyses (UALCAN at https://ualcan.path.uab.edu/ and GEPIA2 at http://gepia2.cancer-pku.cn/#index) of online available datasets from cancer patients (TCGA collection), we generated a list of 3,498 gene whose expression was significantly different (p<0.05) between skin melanoma patients with high- or low-expression of genes which then were ordered according to the value of the S-Ratio calculated as described within the text of the manuscript. According to this experimental design, just as an example, the gene LHB in 1st position of the list has a S-Ratio = 0.3430 which means that patients with higher expression of LHB behave a median survival (1,096 days) about 3 times shorter than patients with lower LHB expression (3,195 days). Similarly, the gene CCL8 in 3,498th position of the list has a S-Ratio = 3,2473 which means that patients with higher expression of CCL8 have a median survival (6,225 days) 3.2 times longer than patients with lower CCL8 expression (1,917 days). Due to the large dimension of such dataset and the complexity of the required analyses, in the present study we focused only on the 25 genes showing the highest and those showing the lowest S-Ratio: a more complete analysis regarding the whole dataset of 3,498 genes is presently ongoing and will be object of a larger investigation.

本研究借助生物信息学(bioinformatics)分析工具UALCAN(https://ualcan.path.uab.edu/)与GEPIA2(http://gepia2.cancer-pku.cn/#index),对公开的癌症患者数据集(癌症基因组图谱The Cancer Genome Atlas,简称TCGA)合集进行分析,得到了3498个基因的列表:在皮肤黑色素瘤患者中,这些基因的高表达组与低表达组之间的表达量存在显著差异(p<0.05);随后根据正文中描述方法计算得到的S-Ratio值对该列表进行排序。按照本次实验设计,举个例子来说,列表首位的LHB基因其S-Ratio值为0.3430:LHB高表达患者的中位生存期为1096天,仅约为低表达患者(3195天)的1/3。同理,列表第3498位的CCL8基因其S-Ratio值为3.2473:CCL8高表达患者的中位生存期为6225天,是低表达患者(1917天)的3.2倍。由于该数据集体量庞大且所需分析流程复杂,本研究仅聚焦于S-Ratio最高与最低的各25个基因;针对全部3498个基因的完整分析目前正在推进中,将作为后续更大规模研究的核心内容。
