Human and animal faeces differ in microbial composition and presence of PCR inhibitors, which affect DNA isolation. While many protocols have been developed for human faeces, not all are equally suitable for animal faecal samples. Here we compared three commercial DNA isolation kits for isolation of DNA from human, cattle, pig and avian (swan) faecal samples: QIAamp FAST DNA Stool Mini Kit (Qiagen), NucleoSpin Soil Mini Kit (Macherey-Nagel) and QIAamp PowerFecal Pro DNA Kit (Qiagen).
人类与动物粪便的微生物组成及聚合酶链式反应(PCR)抑制剂的存在特征均存在差异,此类因素会对DNA提取过程造成影响。尽管目前已针对人类粪便开发出诸多DNA提取方案,但并非所有方案都能同等适用于动物粪便样本。本研究针对人类、牛、猪及鸟类(天鹅)的粪便样本,对比了三款商用DNA提取试剂盒:Qiagen的QIAamp FAST DNA Stool Mini Kit、Macherey-Nagel的NucleoSpin Soil Mini Kit,以及Qiagen的QIAamp PowerFecal Pro DNA Kit。
