Wheat powdery mildew Blumeria graminis forma specialis tritici (B.g. tritici) is a devastating fungal pathogen in agro-ecosystems worldwide and economically the most important powdery mildew disease. The goal of this project is to produce a high quality reference sequence for B.g. tritici. We sequenced the genome of the Swiss isolate 96224 using using next-generation sequencing technology (Roche 454). The assembled sequences were anchored to a backbone of a fingerprint assembly of a B.g. tritici BAC library of the same isolate (FIG 11: 671-677). The resulting reference sequence consists of 251 pseudomolecules. The genome of B.g. tritici has an estimated size of 180Mb and a repetitive content of approximately 90%. The majority of the genes were annotated based on homology to the closely related barley powdery mildew pathogen Blumeria graminis forma specialis hordei (Science 330:1543-1546).

禾本科布氏白粉菌小麦专化型(Blumeria graminis forma specialis tritici,简称B.g. tritici)引发的小麦白粉病,是全球农业生态系统中极具破坏性的真菌病害,也是经济重要性最高的白粉病病害。本项目的目标是为B.g. tritici构建高质量参考基因组序列。研究团队采用下一代测序技术(Roche 454)对瑞士菌株96224开展了全基因组测序。所获组装序列被锚定至该菌株自身的B.g. tritici细菌人工染色体(BAC)文库的指纹图谱组装骨架(图11:671-677)。最终获得的参考序列包含251条假染色体(pseudomolecules)。B.g. tritici的基因组预估大小为180Mb,重复序列占比约90%。绝大多数基因通过与亲缘关系密切的大麦白粉病病原菌禾本科布氏白粉菌大麦专化型(Blumeria graminis forma specialis hordei,Science 330:1543-1546)进行同源性比对完成注释。
