Photographs of leaves inoculated with a Green Fluorescent Protein (GFP) tagged Potato virus Y. Leaves are from 7 accessions of pepper (Capsicum annuum L.), inoculated by aphid transmission (I), mock inoculated (M) or healthy controls (TS). Fluorescence progression was tracked through regular photography at 5, 7, 8, 11, 13, 15, 18, 20, and 22 dpi. Images were captured using a Nikon D5300 digital camera set to aperture F8, ISO 2000, and a shutter speed of 1/2 second, with a picture size of 4496 × 3000 pixels. Fluorescence photographs were captured using a photo bench from the manufacturer BLS (Budapest, Hungary), equipped with lamps designed for GFP excitation, with an emission spectrum ranging from 460 to 495 nm. A green filter with a bandwidth between 500 and 515 nm was mounted on the camera lens to minimize background light and selectively capture GFP emission. A low-intensity white light source was added to allow differentiation of healthy tissues from the background. Focusing was performed under intense white light prior to each photograph; this light was then turned off before the image was captured. The fabric background was selected for its high light-absorption properties to reduce reflections and enhance image clarity.
