RNA-Seq analysis of E15.5 mouse Insm1, Neurod1 and Pax6 knockouts vs Insm1GFP.Cre/+ controls

This experiment used RNA-Seq technology to explore gene expression in mouse Insm1GFP.Cre/+ controls and Insm1 (Insm1GFP.Cre/GFP.Cre), Neurod1 (Insm1GFP.Cre/+; Neurod1LacZ/LacZ), or Pax6 (Insm1GFP.Cre/+; Pax6fl/fl) knockout FACS sorted pancreatic endocrine cells at E15.5. Comparison of Insm1GFP.Cre+/- and knockout animals revealed sets of differentially expressed genes that are required for endocrine cell specification and development.

本实验采用RNA测序(RNA-Seq)技术,探究胚胎期15.5天(E15.5)小鼠的胰腺内分泌细胞的基因表达情况,所使用的细胞分别来自Insm1GFP.Cre/+对照小鼠,以及Insm1基因敲除(Insm1GFP.Cre/GFP.Cre)、Neurod1基因敲除(Insm1GFP.Cre/+; Neurod1LacZ/LacZ)、Pax6基因敲除(Insm1GFP.Cre/+; Pax6fl/fl)小鼠的荧光激活细胞分选(FACS)纯化的胰腺内分泌细胞。通过对比Insm1GFP.Cre+/-杂合对照小鼠与各类基因敲除小鼠的转录组数据,本研究鉴定出一系列参与内分泌细胞特化与发育的必需差异表达基因。
