Intrauterine growth restriction (IUGR) increases the risk of developing type 2 diabetes during adulthood. At day 18 of gestation, we used bilatual uterine artery ligation (BUAL) to restrict nutrient supply to developing rat fetuses to produce IUGR pups. At 2 and 10 weeks of age pancreatic islets were isolated and total RNA extracted using Trizol. Samples with RNA integrity numbers greater than 7 were used to generate libraries
宫内生长受限(intrauterine growth restriction, IUGR)会提升个体成年后罹患2型糖尿病的风险。本研究于妊娠第18天,通过双侧子宫动脉结扎(bilateral uterine artery ligation, BUAL)限制发育中大鼠胎鼠的营养供应,以此构建宫内生长受限幼鼠模型。分别在幼鼠2周龄与10周龄时分离其胰岛,采用Trizol法提取总RNA;选取RNA完整性数大于7的样本构建测序文库。
