Study to characterize the regulatory role of ATRX in the transcriptome of astrocytes and in turn, in brain function. RNA-seq analysis of 1-month-old hippocampus and RNA and ATAC-seq analysis of cortical astrocytic nuclei enriched through sorting were used in a mouse model for targeted deletion of ATRX in astrocytes. Control and ATRX floxed GlastCreER mice were treated with tamoxifen from 10 to 12 days of age. PolyA enriched RNA a was obtained from hole hippocampal tissue. RNA and chromatin were obtained from florescent-labelled cortical astrocytic nuclei.

本研究旨在表征ATRX在星形胶质细胞转录组中的调控作用,进而阐明其对大脑功能的影响。本研究采用星形胶质细胞靶向敲除ATRX的小鼠模型,对1月龄小鼠海马组织开展RNA测序(RNA-seq)分析,并对经分选富集的皮层星形胶质细胞核进行RNA测序(RNA-seq)及转座酶可及性测序(ATAC-seq)分析。对照组及ATRX条件性敲除(floxed)GlastCreER小鼠于出生后10至12日龄接受他莫昔芬(tamoxifen)处理。从完整海马组织中获取聚腺苷酸化(PolyA)富集的RNA样本;从荧光标记的皮层星形胶质细胞核中提取RNA与染色质样本。
