Heart failure (HF) is the ultimate outcome of most cardiovascular disorders with high prevalence and poor prognosis. The transverse aortic constriction (TAC) method is currently the most widely utilized approach for investigating HF induced by left ventricle pressure overload. We explore potential genes with significant changes in HF through single-cell sequencing. Samples were prepared from uninjured mice, as well as from mice which underwent TAC operation 6 weeks to model pressure overload injury. Hearts were dissociated into single cells. We combined single-cell suspensions from 3-5 hearts into one sample.
心力衰竭(Heart Failure, HF)是大多数心血管疾病的终末结局,具有高患病率与不良预后的临床特征。经胸主动脉缩窄(Transverse Aortic Constriction, TAC)法是目前研究左心室压力超负荷诱导型心力衰竭最广泛应用的实验方法。本研究借助单细胞测序(single-cell sequencing)技术,筛选并探索心力衰竭中存在显著表达变化的潜在基因。实验样本分别采集自未造模小鼠,以及接受TAC手术造模6周以构建压力超负荷损伤模型的小鼠。随后将小鼠心脏组织解离为单个细胞,将3-5个小鼠心脏的单细胞悬液合并为一个实验样本。
