Interaction matrices were generated by applying in situ Hi-C to wild-type and deletion mutant trypanosomes. Overall design: DNA-DNA interactions were captured in situ in Trypansoma brucei nuclei, digested with MboI and re-ligated to obtain chimeric DNA molecules of previously interacting chromatin. The ligation products were purified and prepared for paired-end Illumina sequencing. Experiments were performed in duplicates for both the wild-type and the deletion mutant.
本研究通过对野生型(wild-type)与缺失突变体(deletion mutant)布氏锥虫开展原位Hi-C(in situ Hi-C)实验,生成了染色质相互作用矩阵。整体实验设计如下:在布氏锥虫(Trypansoma brucei)的细胞核内原位捕获DNA-DNA相互作用,经MboI酶切后进行再连接,以获取原本存在染色质相互作用的嵌合DNA分子。随后纯化连接产物并制备为双端Illumina测序文库。野生型与缺失突变体两组实验均设置了两次独立重复。
