Treatment of cells by different concentrations of Griffithsin resulted in minor alterations in gene expression Cells were treated with Griffithsin or controls (Cyanovirin-N, lectin deficient Griffithsin termed mGRFT and PBS) and incubated overnight (16 h). Gene expression was compared to that of the PBS treated cells to derive fold changes. Each treatment was done in triplicates. Benjammini and Hochberg correction was used and we considered a fold change of at least 2.0 compared to the PBS treated samples to represent a meaningfully altered gene expression.

采用不同浓度的格里斯福辛(Griffithsin)处理细胞,可引发基因表达的小幅改变。实验将细胞分别用Griffithsin及对照试剂(氰维林-N(Cyanovirin-N)、缺失凝集素结构的Griffithsin突变体mGRFT与磷酸盐缓冲液(PBS))处理,随后孵育过夜(16小时)。以PBS处理组的基因表达水平为参照,计算各组的基因表达倍数变化。每组处理均设置三次生物学重复。本研究采用本杰米尼-霍赫贝格(Benjammini and Hochberg)校正方法,并将相较于PBS处理组的基因表达倍数变化≥2.0,作为判定基因表达发生有意义改变的标准。
